Nikolay Kotov studied in Tomsk drawing classes under the guidance of S. M. Prokhorov. He also studied in Moscow School of painting, sculpture and architecture (MUZhVZ) in 1912–1917, under A. E. Arkhipov, A. M. Vasnetsov, K. A. Korovin and S. D. Miloradovich.

Kotov lived in Tomsk in 1917–1921, since 1921 — in Moscow. He worked as a genre and landscape painter, as poster artist. Kotov was one of the organizers of Tomsk section of Fine Arts. In 1922 he was one of the founders of the Association of the artists of Revolutionary Russia (AKhRR); he took active part in the exhibitions of the Association (1922–1929), including the industrial art exhibition in Cologne in 1929. Kotov showed his works at the exhibition of the Society of artists of Moscow School of painting (1923), the jubilee exhibition Red Army 1918–1923 (1923), the 1st exhibition of the Union of Soviet Artists in Moscow (1931), the 1st travelling art exhibition (Moscow — Saratov — Volgograd — Kazan — Nizhny Novgorod; 1925), and a lot of exhibitions of Moscow Regional Union of Soviet Artists (MOSSKh). The personal exhibition of Kotov was held in Moscow in 1933.

In 1920s–1930s Kotov made several trips to Altai, the Pamirs and Siberia, where he performed a series of paintings and drawings. Kotov drew sketches for panoramas Perekop Sturm (the group of artists; 1934–1941), Tsaritsyn Defence — Stalingrad Defence (together with P. S. Dobrynin; late 1930s — early 1950s). He also took part in reconstruction of the panoramas Borodino Battle and Sevastopol Defence by F. A. Roubaud.

Works by Nikolay Kotov are in many museum collections, including the State Tretyakov Gallery, the State Russian Museum, Tomsk Regional Museum of Local History and others.
